What Does DAO Enzyme Actually Do?
DAO's role is deliberately narrow. It sits mainly in the lining of the small intestine, where it acts on histamine as it arrives from food, breaking it down there before it has the chance to cross into the bloodstream.[1]
This is worth holding onto, because it is the detail most explanations of DAO get wrong. DAO acts on histamine arriving from food. It does nothing about histamine that is released internally by your own mast cells, which is a separate mechanism involved in allergic and inflammatory responses. The two are often conflated in online discussion of histamine intolerance, which muddies the picture for anyone trying to understand their own symptoms, or trying to make sense of the research.
What Foods Are High in Histamine?
Histamine in food is not fixed. It builds up as food ferments, cures or ages. Broadly, the longer and more intensely a food has been fermented or matured, the more histamine it is likely to contain.

Alongside that, there is genuine and growing clinical interest in gut mediated food sensitivity. Searches for terms such as histamine intolerance and low histamine diet have risen steadily, but the quality of what currently ranks for those searches is mixed. A lot of it conflates the two histamine mechanisms, overstates certainty, or skips past practical detail, like food storage, that would actually help someone work out what is going on.
Porcine or Plant: The Two Sources of DAO Supplements
If you look into DAO supplements, you will find two source materials, and they are not equivalent.
Porcine kidney protein extract is DAO derived from pig kidney. This is the form used in the published human trials on histamine intolerance symptom scores, and in the research into migraine and chronic urticaria.[2], [3], [4] The studies are still small and the field is young, but there is a genuine, published evidence base behind this source.
Plant derived DAO, usually from sprouted mung bean or pea, is vegan suitable and commercially appealing for obvious reasons. It has not been studied to anything like the same degree. Research so far has focused on isolating and characterising the enzyme from legume sprouts, and open questions remain around its substrate specificity and how stable its activity is once it is formulated into a supplement.[5]
Why Delivery Format and Timing Matter
DAO is a protein, and stomach acid degrades proteins. That means how a DAO supplement is delivered is not a minor technicality. It is central to whether the enzyme survives long enough to do anything at all.
This also explains why timing shows up so consistently in the research protocols. DAO is taken shortly before a meal, roughly fifteen to twenty minutes beforehand, because the enzyme needs to already be in place in the gut as food arrives. Not with the meal, and not after it. Several of the clinical trials on DAO supplementation used exactly this timing and either one or two capsules taken before eating.[3], [4]
What Is Still Being Established About DAO Deficiency?
It is worth being clear about where DAO sits as a research area. This is not a traditional herbal ingredient with a long history of recorded use. It is a modern, laboratory identified enzyme, and the clinical research into supplementing it is still young. Trials so far have looked at specific, measurable outcomes: symptom scores in histamine intolerance, headache duration in migraine, and activity scores in chronic urticaria.[2], [3], [4]
